I'm thinking of buying a Lenovo Thinkpad X220.

I'm wondering if there is a large difference between 4 GB and 6 GB of memory.

Also, for the 4 GB what is the difference between 1 DIMM and 2 DIMM? Which is better?

4 gigs is fine for average use. If you're using Adobe Photoshop, doing HD video editing or playing demanding 3D games then 6 gigs could be useful. If you get the RAM in 1 DIMM slot, then you'll be able to expand memory by adding a second memory module in the second empty DIMM slot. If you get 4 gigs in 2 DIMM slots (two 2 gig DIMMs, one in each slot), then you'd have to discard one to upgrade memory.

Is there a difference in speed/performance between the 1 DIMM or 2 DIMM choice?

There is no speed difference between 1 and 2 DIMM configurations as long as you use matched DIMMs (same specs for each DIMM) in DIMM pairs. If you buy it with 2 DIMMs, the manufacturer will use matched memory, so there's no need to worry. If you add a second DIMM yourself later, just make sure it's the same type as the first DIMM-- e.g. DDR3 1333MHz in the case of the X220.
